Regulatory Mechanisms of the Calcium Transport System of Fragmented Rabbit Sarcoplasmic Reticulum

چکیده

The outflux of calcium from vesicles of sarcoplasmic reticulum is controlled by the concentration of ionized calcium in the medium. In a calcium-free medium calcium outflux is at a minimum provided ATP and Mg are present. If they are removed calcium outflux becomes rapid and has a rate constant similar to that of outflux during steady-state (40)Ca-(45)Ca exchange with saturating calcium concentrations in the medium. In order to maintain the low permeability state of the membrane only the presence but not the continued hydrolysis of ATP is necessary.

Kinetics of calcium transport by fragmented sarcoplasmic reticulum.

Calcium transport by sarcotubular vesicles (fragmented sarcoplasmic reticulum) from skeletal muscle of rats, rabbits, and humans follows Michaelis-Menten kinetics when studied in systems containing oxalate and low ratios of calcium to vesicle protein. Under these conditions different preparations of vesicles are characterized by their calcium concentrating ability, i.e. the constant level to wh...

Ultrastructure of Sarcoplasmic Reticulum Preparations

Fragmented sarcoplasmic reticulum (FSR) from rabbit muscle was examined by positive staining, negative staining, and freeze-etch electron microscopic techniques in the absence and presence of calcium transport conditions. The existence of 30-40 A particles covering the outer surface of FSR vesicles was confirmed by two different negative stains in unfixed, glutaraldehyde-fixed and osmium tetrox...

Activation of calcium transport in skeletal muscle sarcoplasmic reticulum by monovalent cations.

The rates of calcium transport and Ca2+-dependent ATP hydrolysis by rabbit skeletal muscle sarcoplasmic reticulum were stimulated by monovalent cations. The rate of decomposition of phosphoprotein intermediate of the Ca2+-dependent ATPase of sarcoplasmic reticulum was also increased by these ions to an extent that is sufficient to account for the stimulation of calcium transport and Ca2+-depend...

Effect of physical training on calcium transport by rat cardiac sarcoplasmic reticulum.

Cardiac microsomes enriched in fragmented sarcoplasmic reticulum (SR) were isolated from hearts of physically trained rats and were compared to those from sedentary rats. Preparations from conditioned rats were found to transport calcium to a greater extent than those from sedentary rats both in the absence and in the presence of 1 mM oxalate. Higher oxalate concentrations abolished the differe...

Incorporation of the terminal phosphate of ATP into membranal protein of rabbit cardiac sarcoplasmic reticulum. Correlation with active calcium transport and study of the effects of cyclic AMP.

The transfer of the terminal phosphate of ATP to a material from the sarcoplasmic reticulum of cardiac muscle which can be precipitated by trichloroacetic acid was studied, and the relationship of this biochemical event to active Ca + transport was examined. A component of the phosphoryl transfer reaction was stimulated specifically by Ca + . Both the phosphorylation of the reticulum and the ac...
